Enzyme Processes and Mass Spectrometry

Abstract
A new multiplexing technique for screening combinatorial libraries
of enzyme inhibitors using FTICR and immobilized enzymes will be presented
along with a new method for the calculation of kinetic constants.
Over 1,000 compounds have been screened for several important sulfotransferase
enzymes, including estrogen sulfotransferase. As inhibitors are identified,
they are subjected to a novel strategy for determining Ki kinetic parameters
using an ion trap mass spectrometer. This method allows for the calculation
of Km and Vmax as well as Ki and thus enables the investigator to track
the kinetic mechanisms regarding competitive or non-competitive binding.
Additionally, the non-covalent complexes between substrate/inhibitor and
enzyme are generated using FTICR and structure specificity and folding
characteristics are probed.
Background
Dr. Julie Leary is Professor of Chemistry at UC Berkeley having received
her Ph.D. with Klaus Biemann from MIT. She then had a one year postdoctoral
appointment at Woods Hole Oceanographic and the MIT Mass Spectrometry Resource
Facility before moving to Berkeley. She is recipient of the 2000
Biemann Medal and Sigma Xi awardee. Her current area of research
interest includes carbohdrate analysis from bacteria and pursuit of enzymatic
processes using a multiplexing approach. Julie keeps busy outside
of her career with two teenaged children, a husband and classes in Taiko
to keep her sane.

The San Francisco Bay Area Mass Spectrometry discussion group was formed in 1980 to provide a regular gathering for people interested in mass spectrometry and allied topics. BAMS currently has a membership of about 280 individual and 20 corporate members, and meets 8-10 times per year for a midweek dinner and lecture. Meetings attract between 30 and 90 people, and are held at a restaurant or hotel in the bay area convenient for our speaker. We usually convene at 6:00pm for cocktails, dinner at 7:00pm, and lecture at 8:15pm.
