Faster Drug Optimization: Structural Elucidation and Quantitation of In Vivo Metabolites using Stable Isotopes, Mass Spectrometry and Chemiluminescent Nitrogen Detection

Abstract
In order for a preclinical compound to be developed into a successful
drug, its pharmacokinetic (ADME/PK) properties must be characterized and,
oftentimes, improved. The best method for characterizing ADME/PK properties
is to use a radiolabeled version of the compound, enabling the investigator
to quantitatively track the fate of the compound and its metabolites in
vivo and to determine mass balance. Unfortunately, experiments with radiolabeled
compounds are difficult, expensive, and time consuming. We propose an alternative
that uses a stable label combined with mass spectrometry to trace and identify
the compound and its metabolites, and nitrogen detection to quantitate
them1. In this talk, we will compare the
results of our method to those from a radiolabel study of the same compound.
We will also show a few examples using this method to increase the speed
of the drug optimization process by determining structure-activity relationships
related to distribution, metabolism and excretion as well as validating
in vitro ADME assays.

Background
Gavin received a B.S. in Physics from Yale University and his Ph.D.
in Physics from the University of Illinois, Urbana-Champaign. Following
a postdoctoral fellowship at the University of California at Berkeley,
he joined Cetus Corporation. He is now the director of Analytical Chemistry
in the Small Molecule Drug Discovery Department of Chiron Corporation.
Gavin’s major research interests are the use of biophysical measurements
and mass spectroscopy for the characterization and determination of protein
structure, protein-small molecule affinity and the use of high throughput
tools for the analysis of drugs and their metabolites.

The San Francisco Bay Area Mass Spectrometry discussion group was formed in 1980 to provide a regular gathering for people interested in mass spectrometry and allied topics. BAMS currently has a membership of about 280 individual and 20 corporate members, and meets 8-10 times per year for a midweek dinner and lecture. Meetings attract between 30 and 90 people, and are held at a restaurant or hotel in the bay area convenient for our speaker. We usually convene at 6:00 pm for cocktails, dinner at 7:00 pm, and lecture at 8:15 pm.
