Title: "Mass Spectral Glycomics"
 
ABSTRACT:

Determination of biological phenotype as a function of glycan expression is central to both fundamental cellular processes and applied biomarker development. The fact that glycans are biosynthesized as a distribution of glycoforms on a core structure vastly increases the complexity of the proteome and lipidome in a given biological context. Thus, glycosylation increases both the variability of expressed structures and the biological functions that result from interactions with binding partners. Key to the profiling of released glycan mixtures is a robust and sensitive liquid chromatography LC/MS interface. A chip-based interface will be described for amide-hydrophilic interaction chromatography-MS of released glycans and demonstrated for glycosaminoglycan oligosaccharides. The informatic treatment of glycomics LC/MS datasets differs significantly from that used in proteomics. Glycomics datasets resemble those used in metabolomics and lipidomics in that they comprise a series of structurally related metabolites. The assigning of biological meaning to the data is emerging as the most labor intensive in glycomics mass spectral workflows. A system for using a glycan structural database to query LC/MS datasets generated from heparan sulfate oligosaccharide mixtures will be described. The application of this approach to the comparative profiling of heparan sulfates from different organ tissues will be presented

SPEAKER BIOGRAPHY:

Joseph Zaia is an expert on applications of mass spectrometry in the field of glycomics. He has published on fundamental applied aspects of glycan analysis using tandem mass spectrometry and has developed an LC/MS platform for mass spectral glycomics. He has published a series of highly regarded reviews on this topic. Prof. Zaia received his Ph.D. in organic chemistry from MIT in 1993, working in the laboratory of Prof. Klaus Biemann on peptide sequencing by mass spectrometry. After a one-year post-doctoral fellowship at MIT, he did a second post-doc with Prof. Catherine Fenselau at the University of Maryland, focusing on mass spectrometry of metalloproteins. He joined the cartilage biochemistry group of Osiris Therapeutics, Inc., in June 1996 and worked on the characterization of glycoproteins and proteoglycans from connective tissue. In June 1999 he became Assistant Research Professor of Biochemistry at Boston University School of Medicine. Since this time he has served as Associate Director of the Mass Spectrometry Resource, a NIH/NCRR-funded research resource center. He was promoted to Associate Professor in 2009. He is author or co-author of 46 research publications.

The San Francisco Bay Area Mass Spectrometry discussion group was formed in 1980 to provide a regular gathering for people interested in mass spectrometry and allied topics. BAMS currently has a membership of about 270 individual and 20 corporate members, and meets 7-10 times per year for a midweek dinner and lecture.  Meetings attract between 30 and 90 people, and are held at a restaurant or hotel in the bay area convenient for our speaker.  We usually convene at 6:00 pm for cocktails, dinner at 6:30 to 7:00 pm, and lecture at 7:30 to 8:00 pm.
